Protein Intake and Weight Loss: Underlying Mechanisms

Protein has become one of the most talked-about nutrients. From supermarket shelves filled with high-protein products to social media discussions about protein targets, it is clear that public interest in protein has grown substantially. Much of this attention has focused on its role in building and maintaining muscle, leading many people to associate protein primarily with strength training and athletic performance.
But protein also plays an important role during weight loss. Research suggests that consuming adequate amounts of protein may help increase feelings of fullness, reduce hunger, preserve lean body mass during calorie restriction, and slightly increase the amount of energy the body uses to digest food. Together, these effects may make maintaining a calorie deficit easier while helping preserve muscle during weight loss. (1,2,3).
This article explores what recent human studies tell us about the role of protein intake during weight loss and the mechanisms that may explain its potential benefits.
What does the evidence say?
A 2021 systematic review and meta-analysis that included 43 unique trials, with 37 studies in the body-weight meta-analysis, found that higher-protein diets were associated with modestly greater weight loss than comparison diets. The review also suggested that people with prediabetes may benefit more than those with normoglycaemia, although the authors concluded that the optimal protein intake for weight loss remains uncertain. In a 2011 randomised controlled trial, 27 men with overweight or obesity followed a calorie-restricted diet containing either approximately 0.8 or 1.4 g of protein per kilogram of body weight per day. The higher-protein diet increased fullness throughout the day and reduced late-evening desire to eat and thoughts about food, although it did not significantly reduce average daily hunger. Eating six times per day provided no additional appetite-control benefit, and within the higher-protein group, three eating occasions produced greater evening fullness. These findings suggest that a moderately higher protein intake may help some people find a weight-loss diet easier to stick to by improving satiety and evening appetite control, rather than by requiring people to eat more frequently. In an 8-week randomised intervention from 2022, 121 women with obesity followed a low-energy diet containing different amounts of protein and carbohydrate. Although participants lost an average of approximately 7.5 kg, the higher-protein diets did not produce greater weight loss, satiety, fat loss, or preservation of fat-free mass than the normal-protein diets. However, the groups achieved a smaller difference in protein intake than planned, and the large calorie deficit may have limited the ability to detect more modest effects. These findings suggest that, within a highly restrictive low-energy diet, increasing protein from around 0.9–1.0 to 1.1–1.2 g/kg/day may not provide substantial additional benefits. (5)
In this 6-month randomised controlled trial from 2017, 118 adults with metabolic syndrome followed either a standard-protein diet (0.8 g/kg/day) or a higher-protein diet (1.34 g/kg/day) combined with a 500 kcal/day energy deficit. Both groups lost weight and improved several metabolic health markers. Although the higher-protein group lost more weight numerically, the overall between-group difference in weight loss was not statistically significant after adjustment for baseline BMI, age and sex. However, among participants who closely adhered to the intervention, those consuming the higher-protein diet lost substantially more weight (9.5% vs. 5.8% of baseline body weight). Because the higher-protein group also received protein-enriched meal replacements, the study cannot determine whether these additional benefits were due to the higher protein intake itself, the meal replacements, or better adherence to the dietary programme. What does that mean in practice?
Taken together, the studies suggest that protein is more likely to support the weight-loss process than directly drive weight loss on its own. Much of the potential benefit seems to relate to fullness, appetite control, and how easy the diet is to stick to.
The current evidence is not entirely consistent. While several studies have reported benefits from a higher protein intake, others have found little additional effect, suggesting that factors such as adherence and overall dietary pattern also play a significant role.
A slightly higher protein intake than the current recommended dietary allowance may be beneficial during weight loss, although the optimal amount is still uncertain and is likely to vary between individuals.
Overall, protein should be viewed as an important component of a balanced, calorie-appropriate diet. It is unlikely to compensate for an excessive calorie intake, but it may make the weight-loss process easier to manage.
